Endothelial-Derived Extracellular Vesicles Induce Cerebrovascular Dysfunction in Inflammation

چکیده
Blood–brain barrier (BBB) dysfunction is a key hallmark in the pathology of many neuroinflammatory disorders. Extracellular vesicles (EVs) are lipid membrane-enclosed carriers molecular cargo that involved cell-to-cell communication. Circulating endothelial EVs increased plasma patients with neurological disorders, and immune cell-derived known to modulate cerebrovascular functions. However, little about whether brain cell (BEC)-derived themselves contribute BBB dysfunction. Human cerebral microvascular cells (hCMEC/D3) were treated TNF? IFNy, isolated characterised. The effect on transendothelial resistance (TEER) leukocyte adhesion hCMEC/D3 was measured by electric substrate cell-substrate impedance sensing flow-based T-cell assay. EV-induced changes recipient analysed RT-qPCR Western blotting. A stimulation naïve small (sEVs) reduced TEER shear-resistant adhesion. levels microRNA-155, VCAM1 ICAM1 sEV-treated cells. Blocking expression VCAM1, but not ICAM1, prevented sEV-mediated endothelia. These results suggest sEVs derived from inflamed BECs promote findings may provide new insights into mechanisms involving

Extracellular vesicles derived from Staphylococcus aureus induce atopic dermatitis-like skin inflammation
BACKGROUND Recently, we found that Staphylococcus aureus produces extracellular vesicles (EV) that contain pathogenic proteins. Although S. aureus infection has been linked with atopic dermatitis (AD), the identities of the causative agents from S. aureus are controversial. We evaluated whether S. aureus-derived EV are causally related to the pathogenesis of AD. METHODS Extracellular vesicles...
متن کاملInflammation-induced endothelial cell-derived extracellular vesicles modulate the cellular status of pericytes
Emerging lines of evidence have shown that extracellular vesicles (EVs) mediate cell-to-cell communication by exporting encapsulated materials, such as microRNAs (miRNAs), to target cells. Endothelial cell-derived EVs (E-EVs) are upregulated in circulating blood in different pathological conditions; however, the characteristics and the role of these E-EVs are not yet well understood. In vitro s...
متن کاملExtracellular Vesicles Derived from Gastrointestinal Microbiota: A New Approach to Clinical Studies
Extracellular vesicles, naturally released from all cell types including bacteria, are of great importance in medical microbiology due to transporting a variety of biomaterials, enzymes, and virulence factors, regulating immunity, and having roles in colonization and initiation of signaling pathways. These vesicles are also secreted from microbiota in the gastrointestinal tract and affect the h...
متن کاملGut microbe-derived extracellular vesicles induce insulin resistance, thereby impairing glucose metabolism in skeletal muscle
Gut microbes might influence host metabolic homeostasis and contribute to the pathogenesis of type 2 diabetes (T2D), which is characterized by insulin resistance. Bacteria-derived extracellular vesicles (EVs) have been suggested to be important in the pathogenesis of diseases once believed to be non-infectious. Here, we hypothesize that gut microbe-derived EVs are important in the pathogenesis ...
